Sum it up in a Sentence:
A group of high school girls decide to make a band.
Four girls decide to join an empty light music club at their high-school. One of the characters, Yui, thinks she will be playing basic instruments that kinder-gardeners were taught (which she finds cute). Of course, she misunderstands the idea and later realizes that she is in a rock band. So that the club isn't shut down due to lack of members, she stays in the club and learns to play the electric guitar. Their shyest member, Mio, becomes a back-up singer, as well the as the official song writer. The very enthusiastic member Ritsu becomes the band manager and the drummer. Lastly, Tsumugi becomes the bands piano player. They have several major presentations at their school (such as the school festival). When the band members become sophomores the following year, they allow a freshmen, Azusa, to join the band as well and play the electic guitar with Yui (turns out she is better than Yui).

A very mellow, very chill show about 4 girls starting a high school band through their Light Music Club. No conflict, no problems, no giant mechas destroying the town. Just sweets, tea, and light music.
The thing about K-On! is that it's lack of depth is its strong point. Lack of depth is not something you could normally attribute positively, but it works so well in the series. K-On! follows a group of friends going through everyday life. It's not meant to be dramatic. It's not meant to be anything profound. But that's why it's so good. It has a great flow of comedy, and, if you've ever hung out with a group of friends, then I'm sure you can appreciate the humor in the show.
K-On! just shows those special times in friendship and it does that really well.
